This study explores the effect of new hormone cholesin in the cholesin-GPR146 axis in the cardiovascular system-associated adipose tissue. Our findings reveal that cholesin-GPR146 regulates metabolic pathways and is significantly modulated by the serum lipid levels in the epicardial adipose tissue (EAT). Through collaboration with the cardiosurgical and genetics team at our University (Anna Marcinkiewicz, MD, PhD and prof. Michał Krejca, MD; Izabela Dróżdż, PhD), we analyzed samples from the right cardiac auricle, aortic wall, saphenous vein, and various adipose tissues collected during coronary artery bypass grafting. The study involved 46 participants, with a significant portion having type 2 diabetes. Our RNA sequencing results indicated that GPR146-dependent pathways are enriched with key lipid metabolism regulators, linking this receptor’s activity to the broader lipid profile in patients.
Moreover, our study uncovered that treatment with SGLT2 inhibitors (SGLT2i) inversely affects the GPR146-related gene signature in EAT, suggesting a potential modulatory role of these inhibitors on the cholesin-GPR146 network. This novel insight opens new avenues for understanding how metabolic treatments can impact cardiovascular health at the molecular level.
